Nicotinamide Mononucleotide (NMN) is a nucleotide derived from ribose, nicotinamide, nicotinamide riboside, and niacin (vitamin B3). Its chemical structure is C,,H15N2OP, consisting of a nicotinamide moiety linked to a ribose sugar phosphorylated at the 5‘ position. NMN is the direct precursor to nicotinamide adenine dinucleotide (NAD), a critical coenzyme found in every cell.
Mechanism of Action
NMN is rapidly taken up by cells via the Slc12a8 transporter (in small intestine and other tissues) and converted to NAD* through the salvage pathway, primarily by nicotinamide mononucleotide adenylyl transferases (NMNAT1-3). Elevated NAD* levels activate key NAD+-dependent enzymes:
